Antibody Immunogen Description: Corresponds to amino acid residues S77 to L84. S80 phosphorylation has been linked to inhibition of ACACA enzyme activity. It is one of the major in vivo phosphorylation sites in ACACA (≥ 86 mass spectrometry reports recorded in PhosphoSitePlus. ACACA is known to be phosphorylated at this site in vitro by AMPKa1 (PRKAA1), and AMPKa2 (PRKAA2).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ACACA (acetyl-CoA carboxylase-1; ACC1) is a cytosolic enzyme that catalyzes the carboxylation of acetyl-CoA to malonyl-CoA (ATP + acetyl-CoA + HCO3- <=> ADP + phosphatse + malonyl-CoA), the first and rate-limiting step of de novo fatty acid biosynthesis. The two step reaction starts with the ATP-dependent carboxylation of the biotin carried by the biotin carboxyl carrier (BCC) domain followed by the transfer of the carboxyl group from carboxylated biotin to acetyl-CoA. ACACA is a large, multi-functional enzyme with biotin carboxylase, biotin carboxyl carrier protein, and carboxyltransferase domains. It forms homodimers and homotetramers, which can polymerize into filaments for activation. It is activated by citrate, and inhibited by long-chain fatty acyl-CoAs (feedback inhibition). It is inhibited through phosphorylation by AMPK (AMP-activated protein kinase) when energy levels are low. It is highly expressed in the cytoplasm of lipogenic tissues (liver, adipose).
