Antibody Immunogen Description: Corresponds to amino acid residues H96 to P102. The effect of TP73 Y99 phosphorylation is unclear. This is a minor site of in vivo phosphorylation based on ≥1 mass spectrometry report recorded in PhosphoSitePlus. p73 is known to be phosphorylated at this site in vitro by Abl1.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TP73 (Tumour protein p73) is a member of the p53 family of transcription factors. It acts as a crucial tumour suppressor and regulator of cell stress responses, differentiation, and neurogenesis. It participates in the apoptotic response to DNA damage. It often exists in opposing TAp73 (pro-apoptotic) and delta-Np73 (anti-apoptotic) isoforms, influencing cancer progression and chemosensitivity. Similar to p53, p73 contains a transactivation domain, a DNA-binding domain, and an oligomerization domain. It binds to p53-responsive genes to induce cell cycle arrest and apoptosis. TAp73 contains the N-terminal transactivation domain, promoting apoptosis and acting as a tumour suppressor. Delat-Np73 lacks the N-terminus, acts as an oncogene by inhibiting TAp73 and p53. TP73 is also essential for nervous system development, particularly in the differentiation of multiciliated motile cells. Its activity is regulated by phosphorylation (e.g., by ABL1, PLK1) and degradation through the proteasome. It is expressed in striatal neurons of patients with Huntington disease, and found in rain, kidney, placenta, colon, heart, liver, spleen, skeletal muscle, prostate, thymus and pancreas. It is highly expressed in fetal tissue. p73 is rarely mutated in human cancers, but its expression is dysregulated, with high Delta-Np73/TAp73 ratios often indicating poor prognosis.
