Antibody Immunogen Description: Corresponds to amino acid residues T601 to G608. The effect of SYN1 S605 phosphorylation is unclear. This is a major site of in vivo phosphorylation of SYN1 based on ≥19 mass spectrometry reports recorded in PhosphoSitePlus. SYN1 is known to be phosphorylated at this site in vitro by CaMK2a, PAK1, PAK2, and PAK3 (STK4).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SYN1 (Synapsin I) is a neuron-specific phosphoprotein (approx. 80 kDa) that belongs to the synapsin family; two main forms (synapsin Ia and Ib) are generated by alternative splicing. It is associated with the cytoplasmic surface of synaptic vesicles, and is important for neurotransmitter release and synapse maintenance. It binds vesicles to the actin cytoskeleton in the presynaptic terminal, regulating the reserve pool of vesicles. It functions as a peripheral membrane protein that anchors synaptic vesicles to the cytoskeleton. Upon phosphorylation (e.g., via Protein Kinase A (PKA) or CaMK2), SYN1 releases vesicles, enabling them to move to the plasma membrane for transmitter release. A complex of SYN1 formed with NOS1 and CAPON proteins is necessary for specific nitric-oxid functions at a presynaptic level. It is primarily localized to the presynaptic terminal in neurons.
