Antibody Immunogen Description: Corresponds to amino acid residues F37 to R43. S40 phosphorylation induces activity and transcription. In vivo phosphorylation of NRF2 by high throughput mass spectrometry has not been recorded in PhosphoSitePlus. NRF2 is known to be phosphorylated at this site in vitro by PKCa (PRKCA).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NF2 (Merlin (moesin-ezrin-radixin-like protein); Schwannomin) is a critical tumour suppressor protein. It is a transcription factor which plays a role in the oxidative stress response. It primarily functions in the nervous system, particularly in Schwann cells, to regulate cell proliferation, shape, and adhesion by acting as a link between the cell membrane and the cytoskeleton. Under normal cellular conditions, it is sequestered in the cytoplasm by Keap1 and Cullin3. When under oxidative stress, Nrf2 is release from Keap1 by p62 and is translocated to the nucleus where it acts as a transcription factor for the expression of oxidative stress-related products. NF2 prevents uncontrolled cell growth and division. Inactivation or loss of this protein leads to neurofibromatosis type 2, characterized by benign tumors like schwannomas, meningiomas, and ependymomas.
