Antibody Immunogen Description: Corresponds to amino acid residues S565 to R571. The effect of S568 phosphorylation is unknown. This is the one of the prevalent phosphosite in Nav1.2 (SCN2A) by mass spectrometry (≥6 reports recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Nav1.2 (Sodium channel protein type 2 subunit alpha; SCN2A) is a sodium-selective channel that mediates the voltage-dependent sodium ion permeability of excitable membranes. It belongs to the sodium channel (TC 1.A.1.10) family, Nav1.2/SCN2A subfamily. It is a large, 260 kDa alpha subunit with four repeat domains, each composed of six membrane-spanning segments, often associated with regulatory beta subunits. It takes open or closed conformations in response to the voltage difference across the membrane. The protein forms a sodium-selective channel through which Na+ ions may pass in accordance with their electrochemical gradient. It is suspected to regulate hippocampal replay occurring within sharp wave ripples (SPW-R) important for memory. It is predominantly found on axons and the Axon Initial Segment (AIS) in the brain (cerebral cortex, cerebellum, hippocampus). It is highly expressed in unmyelinated or premyelinated axons and dendrites. Nav1.2 is highly sensitive to blockade by tetrodotoxin (TTX).
