Antibody Immunogen Description: Corresponds to amino acid residues S960 to R966. The effect of KCC2 S963 phosphorylation is unclear. This is one of the major in vivo site of phosphorylation of the KCC2 based on ≥7 mass spectrometry reported recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: KCC2 (Solute carrier family 12 member 5; SLC12A5) is an electroneutral potassium-chloride cotransport protein that belongs to the SLC12A transporter family.It is a neuron-specific, ~130-145 kDa transmembrane protein essential for maintaining low intracellular chloride levels in the central nervous system. It acts as a cotransporter, enabling fast hyperpolarizing inhibition by GABA and glycine. It is crucial for mature neuronal excitability, dendritic spine formation. As major extruder of intracellular chloride, it establishes the low neuronal Cl(-) levels required for chloride influx after binding of GABA-A and glycine to their receptors, with subsequent hyperpolarization and neuronal inhibition. It consists of 12 membrane-spanning segments with intracellular N- and C-terminal domains.
