Antibody Immunogen Description: Corresponds to amino acid residues T601 to G13. The effect of S10 phosphorylation is unclear. This is a minor in vivo site of phosphorylation of H3F3A (≥4 reports from high throughput mass spectrometry studies in PhosphoSitePlus). H3F3A is known to be phosphorylated at this site in vitro by Akt1 (PKBa), AurKB (AurB, STK12), Fyn, IKKa (CHUK, IKBKA), MSK1 (RPS6KA5), MSK2 (RPS6KA4), PKACa (PRKACA), RSK1 (RPS6KA1), RSK2 (RPS6KA3), and SNRK.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: H3F3A (H3-3A, H3.3 histone A) is a replication-independent variant histone protein H3.3, which is incorporated into chromatin to regulate gene expression, particularly at active gene promoters. It constitutes the predominant form of histone H3 in non-dividing cells and is incorporated into chromatin independently of DNA synthesis. H3F3A (H3.3) acts in conjunction with H3F3B to maintain chromatin structure and genome integrity during mammalian development. Mutations in H3F3A, notably K27M and G34R/V, act as "oncohistones" by causing global epigenetic changes that drive specific pediatric-type high-grade gliomas (i.e., K27M) and giant cell tumours of bone (i.e., G34R/V).
