Antibody Immunogen Description: Corresponds to amino acid residues S628 to A635. S632 phosphorylation increases interaction with 14-3-3-beta and 14-3-3-zeta. This is the major site of in vivo phosphorylation with ≥110 mass spectrometry reports recorded in PhosphoSitePlus. HDAC4 is known to be phosphorylated at this site in vitro by PKCA.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: HDAC4 (Histone Deacetylase 4) is a Class IIa zinc-dependent enzyme that primarily functions as a transcriptional repressor by deacetylating cores histones (H2A, H2B, H3 and H4) and non-histone proteins. Removal of acetyl groups from histones causes DNA to become more tightly packed and inaccessible to transcription factors. Beyond histone modification, it acts as a scaffolding protein to modulate the function of various transcription factors (e.g., Runx2, HIF-1α). It deacetylates HSPA1A and HSPA1B at 'Lys-77' leading to their preferential binding to co-chaperone STUB1. It is involved in muscle maturation via its interaction with the myocyte enhancer factors such as MEF2A, MEF2C and MEF2D. HDAC4 contains an N-terminal region for binding transcription factors (e.g., MEF2) and a C-terminal catalytic domain. It suppresses gene expression, particularly by silencing MEF2-dependent target genes. It is essential for chondrocyte hypertrophy in bone formation, muscle fiber differentiation, synaptic plasticity/memory and neuronal survival. It shuttles between the nucleus and cytoplasm. Its localization is regulated by phosphorylation; 14-3-3 protein binding promotes its export from the nucleus to the cytoplasm, inactivating its repressive function.
