Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: eIF4B (Eukaryotic translation initiation factor 4B) is required for the binding of mRNA to the 40S ribosomal subunit of ribosomes. It works in close association with EIF4-F and EIF4-A. It binds near the 5'-terminal cap of mRNA in presence of EIF-4F and ATP, and promotes the ATPase activity and the ATP-dependent RNA unwinding activity of both EIF4-A and EIF4-F. It functions as a multidomain protein with an RNA recognition motif (RRM) and an intrinsically disordered region (IDR) that binds directly to RNA and other initiation factors. It features an N-terminal RNA recognition motif (RRM) and a large, intrinsically disordered C-terminal region (IDR) that supports RNA binding and oligomerization. eIF4B is a key target of the mTOR signaling pathway (via S6K1/2), which phosphorylates the protein at Ser-422 to activate it, especially for mRNAs with complex 5'-UTR structures. Elevated eIF4B activity is linked to cancer progression (e.g., breast cancer, lymphoma) by promoting tumour cell proliferation and survival.
