Antibody Immunogen Description: Corresponds to amino acid residues A67 to T74. S70 phosphorylation of Bcl-2 inhibits its activity. Bcl2 is known to be phosphorylated at this site in vitro by JNK, and JNK3.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Bcl2 (B-cell lymphoma 2) is a 26 kDa integral membrane protein that belongs to the Bcl-2 family, and regulate apoptosis (programmed cell death). It functions as a potent anti-apoptotic protein, crucial for promoting cell survival by preventing mitochondrial outer membrane permeabilization (MOMP), thereby inhibiting the release of cytochrome c and subsequent caspase-mediated cell death. It suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. It functions in a feedback loop system with caspases. and Inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1).
