Antibody Immunogen Description: Corresponds to amino acid residues D361 to D368. S364 and T365 phosphorylation leads to internalization of the receptor. These have not been identified as in vivo phosphorylation sites of TRHR based on mass spectrometry reports recorded in PhosphoSitePlus. TRHR is known to be phosphorylated in vitro at S364 by BARK1 (GRK2, ADRBK1), and at T365 by BARK1 (GRK2, ADRBK1), and CK2a1 (CSNK2A1).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TRHR (Thyrotropin-releasing hormone receptor) is a 7 transmembrane domain G protein-coupled receptor that belongs to the G protein coupled receptor 1 family. It is the receptor for thyrotropin-releasing hormone. This receptor is mediated by G proteins (Gaq and Ga11), which leads to the stimulation of phospholipase C (PLC), resulting in the formation of diacylglycerol (DAG) and inositol triphosphate (IP3), which in turn leads to the release of calcium from calcisomes, and increases cytoplasmic Ca2+.
