Antibody Immunogen Description: Corresponds to amino acid residues R326 to P334. S329 and S331 phosphorylation by PKC-alpha leads to desensitization of the receptor. TBXA2R is known to be phosphorylated in vitro at S329 by PKACa (PRKACA), and PKCa (PRKCA).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TBXA2R (Thromboxane A2 receptor) is a 7-transmembrane, G protein-coupled receptor for thromboxane (TXA2), and belongs to the rhodopsin-like GPCR family (family 1). It is crucial for inducing platelet aggregation, and vasoconstriction of smooth muscles. There are two isoforms of TBXA2R in humans, TPa (Isoform 1) and TPb (Isoform 2), derived from alternative splicing of the TBXA2R gene. The functional activities of this receptor is mediated by G proteins. In the kidney, particularly in glomeruli, arterioles, and mesangial cells, with the binding of TXA2 to glomerular thromboxane prostanoid (TP receptors) induces potent vasoconstriction, leading to reduced glomerular blood flow and lowered glomerular filtration rate (GFR). Activation of the TP receptor by TXA2 or its precursor PGH2 primarily couples with Gq proteins, activating phospholipase C (PLC) and resulting in increased intracellular Ca2+ and protein kinase C (PKC) activation.
