Antibody Immunogen Description: Corresponds to amino acid residues G338 to D346. S341 and S343 phosphorylation leads to desensitization and internalization of the receptor. These have not been reported as in vivo phosphorylation sites in SSTR2 based high throughput mass spectrometry reports in PhosphoSitePlus. SSTR2 is known to be phosphorylated in vitro at S341 by BARK1 (GRK2, ADRBK1), BARK2 (GRK3, ADRBK2), and at S343 by BARK1 (GRK2, ADRBK1), BARK2 (GRK3, ADRBK2), and PKCa (PRKCA).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SSTR2 (Somatostatin receptor 2) is a 7-transmembrane G protein-coupled receptor that acts as the primary, highly expressed target for somatostatin-14 and -28 in neuroendocrine cells. It inhibits hormone release (GH, insulin) and cell proliferation. It is a critical diagnostic and therapeutic target for neuroendocrine tumours (NETs). There are two main isoforms (A and B). It inhibits adenylyl cyclase and reduces calcium-dependent exocytosis, leading to decreased hormone secretion. This receptor is coupled via pertussis toxin sensitive G proteins to inhibition of adenylyl cyclase. In addition it stimulates phosphotyrosine phosphatase and PLC via pertussis toxin insensitive as well as sensitive G proteins. It inhibits calcium entry by suppressing voltage-dependent calcium channels. It inhibits cell growth through enhancement of MAPK1 and MAPK2 phosphorylation and subsequent up-regulation of CDKN1B.
