Antibody Immunogen Description: Corresponds to amino acid residues A348 to D356. S352 and S354 phosphorylation leads to desensitization of the receptor by promoting its internalization. These are two of the major in vivo phosphorylation sites in P2RY1 by mass spectrometry (≥2 and ≥2 reports, respectively, in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: P2RY1 (P2Y purinoceptor 1) is a 7-transmembrane G protein-coupled receptor (7TM GPCR) of the Type 1 G protein-coupled receptor family. It functions as a major receptor for extracellular adenosine diphosphate (ADP) and adenosine triphosphate (ATP). The receptor activates Gq class G proteins, leading to the stimulation of phospholipase C (PLC) and the mobilization of intracellular calcium ions (Ca2+). It also regulates the PI3K/Akt/GSK3 axis depending in astrocytes and neural cells, and in platelets. Alongside P2RY12, P2RY1 is crucial for ADP-induced platelet shape change and aggregation. It also mediates cellular processes such as calcium signalling, proliferation, endothelial cell migration, alveolar bone resorption, purinergic signaling in the nervous system, thrombosis and apoptosis. In astrocytes, P2RY1 signalling inhibits Glycogen Synthase Kinase-3 beta (GSK3b) by promoting its phosphorylation at its inhibitory Ser- 9 site in a PKC-dependent manner that does not require Akt.
