Antibody Immunogen Description: Corresponds to amino acid residues E306 to A312. T308 and S310 phosphorylation is inhibitory to downstream signalling by inducing receptor desensitization. These are minor in vivo phosphorylation sites in LTB4R (≥2 and ≥2 reports, respectively, from high throughput mass spectrometry studies recorded in PhosphoSitePlus). LTB4R is known to be phosphorylated in vitro at T308 by GRK6 (GPRK6).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: LTB4R (Leukotriene B4 Receptor 1; BLT1) is a 7-transmembrane G protein-coupled receptor (7TM GPCR) of the G protein-coupled receptor 1 family. It is a high affinity receptor for Leukotriene B4, and extracellular ATP > UTP and ADP. It is primarily expressed on leukocytes, and also acts as a high-affinity receptor for the potent chemoattractant leukotriene LTB4. The activity of this receptor is mediated by pertusissis toxin-senstive Gi-like G proteins which activate a phosphatidylinositol C-calcium second messenger system. Via activation of MAPK's and NF-kappa B, it triggers inflammatory responses, including chemotactic migration of neutrophils, macrophages, and leukocytes to sites of tissue injury and infection, leukocyte activation, and cytokine production. It may also be the cardiac P2Y receptor involved in the regulation of cardiac muscle contraction through modulation of L-type calcium currents. It is Implicated in inflammatory diseases, including asthma, arthritis and atherosclerosis, psoriasis, and cancer progression (e.g., renal cell carcinoma).
