Antibody Immunogen Description: Corresponds to amino acid residues D366 to D372. Y369 phosphorylation (which is a serine residue in mouse) may lead to desensitization of the receptor to its agonist based on mouse studies. This is a minor in vivo phosphorylation site in KOR1 (1 report from high throughput mass spectrometry studies recorded in PhosphoSitePlus). KOR-1 is known to be phosphorylated at this site in vitro by BARK2 (GRK3 ADRBK2), GRK5 (GPRK5), and GRK6 (GPRK6).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: KOR1 (Kappa-opioid receptor; OPRK1) is a 7-transmembrane G protein-coupled receptor (7TM GPCR) that belongs to the G protein-coupled receptor (GPCR) 1 family. It plays a critical role in mediating pain perception, mood regulation, reward systems, stress responses, and motor control. Besides endogenous dynorphins (specifically dynorphin A), KOR-1 acts as a receptor for synthetic opioids and the psychoactive diterpene salvinorin A. It has a low affinity for beta-endorphins. Upon binding ligands, KOR-1 triggers intracellular signaling, typically coupling to inhibitory Gi/Go proteins. This inhibits adenylate cyclase activity, reduces calcium ion currents, and increases potassium ion conductance, leading to reduced neurotransmitter release. KOR activation is associated with analgesia (pain relief), as well as the induction of dysphoria, anxiety, and depression, which is distinct from mu-opioid receptor activation. It is also involved in regulating body temperature, water balance, and neuroendocrine functions. Down-regulation of KOR-1 may promote cancer progression, specifically in esophageal squamous cell carcinoma, by enhancing proliferation, migration, and invasion via the PI3K-Akt signaling pathway.
