Antibody Immunogen Description: Corresponds to amino acid residues L4264 to R4270. The effect of S4267 phosphorylation is unclear. This is the major in vivo phosphorylation site in KMT2C based on ≥52 mass spectrometry reports recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: KMT2C (Lysine Methyltransferase 2C; MLL3) is a nuclear histone methyltransferase that belongs in the histone-lysine methyltransferase family, TRX/MLL subfamily. It adds methyl groups to the Lys-4 position of histone H3 (H3K4me1/2), which activates enhancers and facilitates gene transcription. It catalyzes the reaction: L-lysyl4-[histone H3] + S-adenosyl-L-methionine = N6-methyl-L-lysyl4-[histone H3] + S-adenosyl-L-homocysteine + H+. It regulates gene expression, differentiation, and DNA repair within the COMPASS-like complex. It works closely with its paralog, KMT2D (MLL4). It features a SET domain (catalytic activity), PHD-type zinc fingers, and a RING-type zinc finger. It acts as a tumour suppressor by mediating histone H3 lysine 4 (H3K4) mono- and di-methylation at enhancer regions. Loss-of-function mutations in KMT2C are frequently found in various cancers (e.g., gastric, prostate, leukemia). It is also associated with neurodevelopmental disorders such Kleefstra syndrome-2. It is highly expressed in testis and ovary, followed by brain and liver. It is also expressed in placenta, peripherical blood, fetal thymus, heart, lung and kidney. Within brain, expression was highest in hippocampus, caudate nucleus, and substantia nigra. It does not appear to be detectable in skeletal muscle and fetal liver.
