Antibody Immunogen Description: Corresponds to amino acid residues E355 to G367. T358, T361 and S363 phosphorylation of the S363 site by GRK2 leads to desensitization of the receptor to its agonist and internalization of the receptor. These are minor in vivo phosphorylation sites in DOR1 (≥0, ≥1 and ≥1 reports, respectively, from high throughput mass spectrometry studies recorded in PhosphoSitePlus). DOR1 is known to be phosphorylated in vitro at T361 by GRK2, GRK3. S363: GRK2, and GRK3.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: DOR1 (Delta-type opioid receptor 1; OPRD1) is a 30.5 kDa 7-transmembrane G protein-coupled receptor (7TM GPCR) that belongs to the G protein-coupled receptor 1 family. It functions as receptor for endogenous enkephalins and for a subset of other opioids. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (Gi/Go proteins) and modulates the activity of down-stream effectors, such as adenylate cyclase. DOR1 signalling leads to the inhibition of adenylate cyclase activity. It inhibits neurotransmitter release by reducing calcium ion currents and increasing potassium ion conductance. It functions in the perception of pain and in opiate-mediated analgesia, and plays a role in developing analgesic tolerance to morphine. It is expressed in the plasma membrane and vesicles of neurons, particularly in the brain's basal ganglia and neocortical regions (cerebral cortex), hypothalamus, hippocampus, olfactory bulb and oocytes.
