Antibody Immunogen Description: Corresponds to amino acid residues A73 to S78. The effect of Y74 phosphorylation is unclear. This is a major site of in vivo phosphorylation with 52 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: DARPP32 (Protein phosphatase 1 regulatory subunit 1B; PPP1R1B) belongs to the protein phosphatase inhibitor 1 family. It mediates dopamine, glutamate, and other neurotransmitter signals in medium-size spiny neurons, regulating responses to antipsychotics and drugs of abuse. It serves as a central hub, integrating signals from various neurotransmitters (dopamine, glutamate, serotonin) and mediating actions of stimulants like cocaine, nicotine, and caffeine. It is a potent inhibitor of protein phosphatase-1 (PP1) when phosphorylated by protein kinase A (PKA) at Thr-34. It has also been implicated as a PKA inhibitor when phosphorylated at Thr-75 by CDK5. DARPP32 activity is further modulated by phosphorylation at other sites (Ser-97, Ser-102, Ser-130) by kinases such as CK1 and CK2, which regulate its subcellular localization.
