Antibody Immunogen Description: Corresponds to amino acid residues G450 to Q459. S451, T455 and S457 phosphorylation is likely to be inhibitory to downstream signalling. These are minor in vivo phosphorylation sites in Chk2 (≥4, ≥3 and ≥3 reports, respectively from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CHRM1 (Muscarinic acetylcholine receptor M1; mAChR m1) is a 51 kDa 7-transmembrane domain receptor that belongs to the G protein-coupled receptor (1 family (7TM GPCR), Muscarinic acetylcholine receptor subfamily, CHRM1 sub-subfamily. It binds the neurotransmitter acetylcholine to modulate neuronal activity, memory consolidation, cognitive processing, and synaptic plasticity. Downstream signalling includes inhibition of adenylate cyclase, breakdown of phosphoinositides and modulation of potassium channels through the action of G proteins. Its primary transducing effect is Pi turnover. It is mainly expressed in the brain (cortex, hippocampus), but also involved in vagally induced bronchoconstriction. It is essential for regulating REM sleep, often acting in concert with CHRM3. Reduced expression of CHRM1 is linked to schizophrenia.
