Antibody Immunogen Description: Corresponds to amino acid residues R67 to Q73. The effect of S70 phosphorylation is unclear. This is a major site of phosphorylation for BRCA2 (≥14 reports by mass spectrometry in PhosphositePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: BRCA2 (Breast cancer type 2 susceptibility protein) is a DNA repair enzyme and nuclear receptor co-regulator. It is involved in double-strand break repair and/or homologous recombination. It binds RAD51 and potentiates recombinational DNA repair by promoting assembly of RAD51 onto single-stranded DNA (ssDNA). It acts by targeting RAD51 to ssDNA over double-stranded DNA, enabling RAD51 to displace replication protein-A (RPA) from ssDNA and stabilizing RAD51-ssDNA filaments by blocking ATP hydrolysis. It is part of a PALB2-scaffolded HR complex containing RAD51C and thought to function in DNA repair by HR. It may participate in S phase checkpoint activation. It bnds selectively to ssDNA, and to ssDNA in tailed duplexes and replication fork structures. It may play a role in the extension step after strand invasion at replication-dependent DNA double-strand breaks; together with PALB2, BRCA2 is involved in both POLH localization at collapsed replication forks and DNA polymerization activity. In concert with NPM1, it regulates centrosome duplication. It interacts with the TREX-2 complex (transcription and export complex 2) subunits PCID2 and SEM1, and is required to prevent R-loop-associated DNA damage and thus transcription-associated genomic instability. Silencing of BRCA2 promotes R-loop accumulation at actively transcribed genes in replicating and non-replicating cells, indicating that BRCA2 mediates the control of R-loop associated genomic instability, independently of its known role in homologous recombination.
