Antibody Immunogen Description: Corresponds to amino acid residues M363 to R371. S366 and T369 phosphorylation is likely to be inhibitory to downstream signalling. BKR2 is known to be phosphorylated in vitro at S366: by GRK4.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: BKR2 (B2 bradykinin receptor) is a 7-transmembrane G protein-coupled receptor (7TM GPCR) that belongs to the G protein-coupled receptor Type 1 family, Bradykinin receptor subfamily. Bradykinin (released during trauma and inflammation) and kallidin (lys-bradykinin) act as ligands for this receptor. It is associated with G proteins that activate a phosphatidylinositol-calcium second messenger system. Upon ligand binding, BKR2 activates Gq and Gi proteins, resulting in the stimulation of phospholipase C (PLC) and an increase in intracellular free calcium. It also activates the mitogen-activated protein kinase (MAPK) and phosphatidylinositol 3-kinase (PI3K) pathways.
