Antibody Immunogen Description: Corresponds to amino acid residues K375 to S382. T378 and S380 phosphorylation is likely to be inhibitory to downstream signalling. The T378 phosphorylation has not been detected by mass spectrometry and the S380 site has been reported only once in PhosphoSite Plus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: AVPR1A (Vasopressin V1a receptor) is a 7-transmembrane G protein-coupled receptor (7TM GPCR) that belongs to the G protein-coupled receptor 1 family, Vasopressin/oxytocin receptor subfamily. It is a receptor for arginine vasopressin. The activity of this receptor is mediated by G proteins (e.g., Gaq, Ga11) which activate a phosphatidyl-inositol-calcium second messenger system. It has been linked to the manifestation of social behaviors, including affiliation and attachment. It regulates cardiovascular homeostasis, cell proliferation, and hepatic glycogenolysis. It is highly expressed in vascular smooth muscle, liver, and various regions of the brain. Differences in regional receptor expression in the brain as well as differences in social behavior may result from a highly variable repetitive sequence in the 5' flanking region of AVPR1A. One such allelic variant has been linked to autism.
