Antibody Immunogen Description: Corresponds to amino acid residues T332 to P341. S335, T336 and S338 phosphorylation is likely to be inhibitory to downstream signalling, although there are resports of stimulated activity. This is likely targeted by a GRK family kinase.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Its ligand is angiotensin II, a vasoconstricting peptide, which acts as a key regulator of blood pressure and sodium retention by the kidney. The ligand-activated AGTR1 couples to G-alpha proteins G(q) (GNAQ, GNA11, GNA14 or GNA15) and stimulates phospholipase C to indirectly increase the cytosolic Ca2+ concentrations, which in turn triggers cellular responses such as stimulation of protein kinase C-alpha and beta isoforms. Mutations can cause renal tubular dysgenesis, a severe disorder of kidney development. It is a key target for antihypertensive medications such as Losartan, Valsartan, and Candesartan, which act as AT1 receptor blockers.
