Antibody Immunogen Description: Corresponds to amino acid residues G351 to N359. S355 and S356 phosphorylation is inhibitory to some downstream signalling by promoting desensitization, binding to beta-arrestin (ARRB2) and receptor internalization and degradation. ADRB2 is known to be phosphorylated at this site in vitro by BARK1 (GRK2, ADRBK1), GRK3, and GPRK5 (GRK5).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ADRB2 (beta-2 adrenergic receptor) is a 7-transmembrane G protein-coupled receptor (7TM GPCR) for catecholamines that couples to both G(s) and G(i) proteins, activating bifurcated signalling pathways. It belongs to the G protein-coupled receptor 1 family. Adrenergic receptor subfamily. ADRB2 sub-subfamily. It binds epinephrine (Epi) with an approximately 30-fold greater affinity than norepinephrine (NE). Both NE and Epi promote coupling to G(s)/PKA pathway to regulate myocyte contraction rate. Epi also promotes ADRB2 coupling to G(i) proteins to exert cardioprotective effects especially in the conditions of hypoxia and oxidative stress through the G(i)/PI3K/Akt signalling pathway. In the heart, Epi- and NE-activated ADRB2 are thought to induced rapid and slow cardiomyocyte contraction rates, respectively. ADRB2-G(s) signalling delivers proapoptotic signals in cardiomyocytes, whereas G(i)-mediated survival effects predominate.
