Antibody Immunogen Description: Corresponds to amino acid residues R108 to G114. S111 phosphorylation induces interaction with Tip60 and p300. This is a minor site of in vivo phosphorylation of VHL based on ≥1 mass spectrometry reports recorded in PhosphoSitePlus. VHL is known to be phosphorylated at this site in vitro by Chk2.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: VHL (von Hippel-Lindau) protein is a crucial tumour suppressor in the VHL family. It functions as a target recruitment subunit in the E3 ubiquitin ligase complex, and regulates oxygen sensing and cell proliferation. It recruits hydroxylated hypoxia-inducible factor (HIF-1-alpha) for proteasomal degradation under normal oxygen levels, preventing tumour growth. The VHL protein helps stop cells from growing and dividing too rapidly, serving as a critical "gatekeeper" in cancer prevention. VHL acts as the substrate-recognition subunit of the VCB-CUL2 complex (comprising VHL, Elongin B, Elongin C, and Cullin-2). By tagging HIF-1 for degradation in oxygen-rich environments, it prevents improper angiogenesis (blood vessel formation) and metabolic reprogramming.
