Antibody Immunogen Description: Corresponds to amino acid residues S349 to K355. The effect of Y352 phosphorylation is unclear, but it is the major site of UGDH phosphorylation by mass spectometry studies (≥63 reports recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: UDP-glucose dehydrogenase (UGDH) is a cytosolic and nuclear hexameric enzyme that belongs to the UDP-glucose/GDP-mannose dehydrogenase family. It converts UDP-glucose to UDP-glucuronic acid (UDP-GlcUA) using NAD+, and it is important for producing extracellular matrix components like hyaluronan. It plays a vital role in glycosaminoglycan synthesis, xenobiotic metabolism, and cancer progression. It catalyzes the two-step oxidation of UDP-glucose to UDP-glucuronic acid, which is a precursor for glycosaminoglycans (hyaluronan, chondroitin sulfate). As a homohexamer (dimer-of-trimers) it undergoes conformational changes during catalysis. It enzymatic activity is regulated by allosteric feedback, such as by UDP-xylose. It is expressed in heart, placenta, liver, pancreas, spleen, thymus, prostate, ovary, small intestine and colon.
