Antibody Immunogen Description: Corresponds to amino acid residues G501 to Y508. The effect of S505 phosphorylation is unclear. This is the major site of in vivo phosphorylation of TSC1 based on ≥86 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TSC1 (Hamartin) is a tumour suppressor protein that forms a complex with Tuberin (TSC2) and TBC1D7. This TSC complex inhibits mTORC1, a major regulator of anabolic growth, and functions as a co-chaperone for Hsp90 to stabilize TSC2. TSC1 is essential for controlling cell growth and metabolic activity, and its deficiency leads to overactive mTORC1. It interacts directly with TSC2 (tuberin) to form the TSC complex, which functions as a GTPase-activating protein (GAP) for the Rheb GTPase, reducing mTORC1 activity. It inhibits the nutrient-mediated or growth factor-stimulated phosphorylation of S6K1 and EIF4EBP1 by negatively regulating mTORC1 signaling. TSC1 acts as a co-chaperone for Hsp90, preventing the degradation of TSC2 and facilitating its stabilization, as well as chaperoning of protein clients such as kinases, TSC2 and glucocorticoid receptor NR3C1. TSC1 increases ATP binding to HSP90AA1 and inhibits HSP90AA1 ATPase activity. It competes with the activating co-chaperone AHSA1 for binding to HSP90AA1, thereby providing a reciprocal regulatory mechanism for chaperoning of client proteins. It recruits TSC2 to HSP90AA1 and stabilizes TSC2 by preventing the interaction between TSC2 and ubiquitin ligase HERC1. It is involved in microtubule-mediated protein transport, but this seems to be due to unregulated mTOR signaling.It is highly expressed in skeletal muscle, followed by heart, brain, placenta, pancreas, lung, liver and kidney. Also expressed in embryonic kidney cells.
