Antibody Immunogen Description: Corresponds to amino acid residues K222 to P231. The effects of T225 and S228 phosphorylation are unclear. These are minor sites of in vivo phosphorylation of TPH1 with only 1 mass spectrometry report for each site in PhosphoSitePlus. However, there are very few phosphosites in this protein and they are only reported once.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TPH1 (Tryptophan Hydroxylase 1) is a 50.9 kDa rate-limiting enzyme that catalyzes the crucial first and rate limiting step in the biosynthesis of peripheral serotonin, which is later made into melatonin in the pineal gland. As a Fe2+-dependent homotetramer, this oxidoreductase specifically converts L-tryptophan to 5-hydroxy-L-tryptophan (5-HTP) in the reaction: (6R)-L-erythro-5,6,7,8-tetrahydrobiopterin + L-tryptophan + O2 = 5-hydroxy-L-tryptophan + (4aS,6R)-4a-hydroxy-L-erythro-5,6,7,8-tetrahydrobiopterin. It belongs to the biopterin-dependent aromatic amino acid hydroxylase family. While TPH1 is responsible for peripheral serotonin production, its homolog, TPH2, is responsible for central serotonin synthesis in the brain. TPH1 is primarily expressed in the gut, pineal gland, and mast cells, and regulates serotonin levels for various physiological functions, including GI motility and immune response. Isoform 1 is more widely expressed than isoform 2.
