Antibody Immunogen Description: Corresponds to amino acid residues T273 to A280. S276 phosphorylation induces transcription and interaction with HDAC1 and SIN3A. This is the major site of in vivo phosphorylation of RUNX1 (AML1) based on ≥76 mass spectrometry reports recorded in PhosphoSitePlus. RUNX1 is known to be phosphorylated at this site in vitro by CDK1 (CDC2), CDK2, CDK6, ERK1 (MAPK3), ERK2 (MAPK1), and HIPK2.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: RUNX1 (Runt-related transcription factor 1; AML1) is a critical transcription factor protein that regulates gene expression for hematopoietic stem cell differentiation into mature blood cells. It forms the heterodimeric complex core-binding factor (CBF) with CBFB, which stabilizes it and enhances its DNA-binding capacity. The CBF complex, binds to DNA to regulate hematopoiesis. It is important for establishing definitive hematopoiesis (blood cell formation) and immune system development. Beyond blood formation, it functions in nerve development (specifically pain sensing) and has roles in inflammatory responses, including the pathway. RUNX members modulate the transcription of their target genes through recognizing the core consensus binding sequence 5'-TGTGGT-3', or very rarely, 5'-TGCGGT-3', within their regulatory regions via their runt domain, while CBFB is a non-DNA-binding regulatory subunit that allosterically enhances the sequence-specific DNA-binding capacity of RUNX. The heterodimer complex bind to the core site of a number of enhancers and promoters, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, LCK, IL3 and GM-CSF promoters.
