Antibody Immunogen Description: Corresponds to amino acid residues R772 to G779. S775 phosphorylation stimulates enzymatic activity. This is the major in vivo phosphorylation site in PFKL (≥68 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PFKL (ATP-dependent 6-phosphofructokinase, liver type) belongs to the phosphofructokinase type A (PFKA) family. ATP-dependent PFK group I subfamily. Eukaryotic two domain clade E sub-subfamily. It catalyzes the phosphorylation of D-fructose 6-phosphate to fructose 1,6-bisphosphate by ATP, the first committing step of glycolysis. It negatively regulates the phagocyte oxidative burst in response to bacterial infection by controlling cellular NADPH biosynthesis and NADPH oxidase-derived reactive oxygen species. In activated macrophage, PFKL drives the metabolic switch toward glycolysis, and prevents glucose turnover for NADPH production via pentose phosphate pathway. PFKL inhibition, such as via O-GlcNAcylation, can shift glucose metabolism toward the pentose phosphate pathway for antioxidant defense. As a homotetrameric regulatory protein, PFKL plays a critical role in cellular energy metabolism, specifically within the liver, kidneys, and leukocytes.
