Target Alias: 6PF-2-K/Fru-2,6-P2ase brain/placenta-type isozyme; 6-phosphofructo-2-kinase; F263; Fructose-2,6-bisphosphatase; IPFK-2; PFK-2/FBPase-2, brain/placenta-type; PFKFB-3

Product Type Specific: PFKFB3 phosphosite-specific antibody

Antibody Code: PN769

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: S467

Protein UniProt: Q16875

Protein SigNET: PFKFB3

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Synthetic phosphopeptide patterned after human PFKFB3

Antibody Immunogen Sequence: PLA(pS)PEP(βA)C

Antibody Immunogen Description: Corresponds to amino acid residues P464 to P470. S467 phosphorylation induces cell growth. This is one of the major in vivo phosphorylation sites by mass spectrometry (≥56 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.25 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Rhesus macaque | Rat | Mouse | Zebra fish.

Scientific Background: PFKFB3 (6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3) is a bifunctional enzyme that belongs to the phosphoglycerate mutase family. It is a key metabolic regulator that synthesizes fructose-2,6-bisphosphate (F2,6BP), a potent activator of glycolysis. It serves as a, "master regulator" of glycolysis. It has high kinase activity to produce F2,6BP, which activates 6-phosphofructo-1-kinase (PFK-1), the rate-limiting enzyme of glycolysis. Beyond glycolysis, PFKFB3 can localize to the nucleus, where it activates Cyclin-dependent kinase 1 (Cdk1) and regulates cell cycle progression. It exists in several isoforms, with the carboxyl-terminal domain influencing its localization and activity. It is highly expressed in solid tumours, leukemia, and rapidly proliferating cells. It promotes high glycolytic flux, cell proliferation, and survival. It sustains the high glycolytic rate characteristic of the Warburg effect in cancer cells, aiding tumour growth and angiogenesis. Its inhibition leads to apoptosis and cell cycle arrest. This description may include information annotated by UniProt and/or Google AI.
