Antibody Immunogen Description: Corresponds to amino acid residues P464 to P470. S467 phosphorylation induces cell growth. This is one of the major in vivo phosphorylation sites by mass spectrometry (≥56 reports recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PFKFB3 (6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3) is a bifunctional enzyme that belongs to the phosphoglycerate mutase family. It is a key metabolic regulator that synthesizes fructose-2,6-bisphosphate (F2,6BP), a potent activator of glycolysis. It serves as a, "master regulator" of glycolysis. It has high kinase activity to produce F2,6BP, which activates 6-phosphofructo-1-kinase (PFK-1), the rate-limiting enzyme of glycolysis. Beyond glycolysis, PFKFB3 can localize to the nucleus, where it activates Cyclin-dependent kinase 1 (Cdk1) and regulates cell cycle progression.
