Antibody Immunogen Description: Corresponds to amino acid residues R300 to A307. The effect of S303 phosphorylation is unclear. This is the major site of in vivo phosphorylation of PBRM1 by mass spectrometry (≥105 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PBRM1 (Protein polybromo-1; BAF180) is a 193 kDa nuclear protein that acts as a key chromatin-targeting subunit of the PBAF (Polybromo-associated BAF) chromatin remodeling complex. It features six tandem bromodomains (BD1–6), two bromo-adjacent homology (BAH) domains, and a C-terminal high mobility group (HMG) box, which collectively allow it to recognize acetylated histone marks and remodel chromatin to regulate gene expression. The bromodomains act as "readers" of acetylated histone lysine residues, while the BAH domains are involved in binding unmodified histones H3 and H4. As a defining component of the PBAF complex (a subset of SWI/SNF), PBRM1 uses ATP to alter DNA-nucleosome structure for transcriptional activation or repression. Besides chromatin remodeling, it regulates cell proliferation and can bind to double-stranded RNA via its bromodomains to facilitate chromatin interaction. It is widely expressed in tissues. PBRM1 is a well-known tumour suppressor frequently mutated in ~40–50% of clear cell renal cell carcinomas (ccRCC), with significant mutations also found in lung, and breast cancers.
