Antibody Immunogen Description: Corresponds to amino acid residues A90 to L97. The effect of ODC1 T93 phosphorylation is unclear. This is one of the few in vivo phosphorylation sites of ODC1 that has been identified by mass spectrometry (≥2 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ODC1 (Ornithine decarboxylase 1) is a 48-53.5 kDa protein that belongs to the Orn/Lys/Arg decarboxylase class-II family. It is the first and rate-limiting enzyme in polyamine biosynthesis with its conversion of ornithine to putrescine with the enzyme reaction: L-ornithine + H+ = putrescine + CO2. Polyamines, such as putrescine, spermidine, and spermine, are essential for cell proliferation and are implicated in cellular processes, ranging from DNA replication to apoptosis. It is highly expressed in rapidly growing tissues. Its activity is tightly regulated by ubiquitin-dependent and independent degradation. It is inhibited by S-nitrosylation. It is inhibited by antizymes (AZs) OAZ1, OAZ2 and OAZ3 in response to polyamine levels. AZs inhibit the assembly of the functional homodimer by binding to ODC monomers. Additionally, OAZ1 targets ODC monomers for ubiquitin-independent proteolytic destruction by the 26S proteasome. At least 13 ubiquitinylated sites have been identified in ODC1. It is often upregulated in cancers (e.g., neuroblastoma, prostate, colon) due to oncogenes like MYCN/c-Myc. It is down-regulated in response to enterovirus 71 (EV71) infection.
