Antibody Immunogen Description: Corresponds to amino acid residues N148 to F154. Y151 phosphorylation is inhibitory. In vivo phosphorylation of the Y151 site in NOS2 has not been reported in high throughput mass spectrometry studies recorded in PhosphoSitePlus. NOS2 is known to be phosphorylated at this site in vitro by Src.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NOS2 (Nitric oxide synthase 2; iNOS) is an inducible, calcium-independent enzyme that generates large amounts of nitric oxide (NO) for immune defense and inflammatory responses. NO mediates tumouricidal and bactericidal actions. Unlike constitutive isoforms (NOS1, NOS3), NOS2 expression is induced by inflammatory stimuli, such as cytokines (TNF-alpha, IL-1-beta) and bacterial lipopolysaccharide (LPS). As a 1153-amino acid, heme-containing homodimer, it catalyzes the conversion of L-arginine to L-citrulline and NO. In macrophages, it functions as an oxidoreductase, using cofactors like NADPH, FAD, FMN, and tetrahydrobiopterin (BH4) to produce nitric oxide. The catalyzed reaction is: 2 L-arginine + 3 NADPH + 4 O2 + H+ = 2 L-citrulline + 2 nitric oxide + 3 NADP+ + 4 H2O. It also has nitrosylase activity and mediates cysteine S-nitrosylation of cytoplasmic target proteins such PTGS2/COX2. It features a C-terminal reductase domain and an N-terminal oxygenase domain. It is a component of the iNOS-S100A8/9 transnitrosylase complex involved in the selective inflammatory stimulus-dependent S-nitrosylation of GAPDH on 'Cys-247' implicated in regulation of the GAIT complex activity, and probably multiple targets including ANXA5, EZR, MSN and VIM. It is involved in inflammation, enhances the synthesis of proinflammatory mediators such as IL6 and IL8. It is expressed in the liver, retina, bone cells and airway epithelial cells of the lung, but not in platelets. While protective against pathogens (viruses, bacteria, fungi), excessive NOS2-derived NO is associated with inflammatory diseases, sepsis, and cancer.
