Antibody Immunogen Description: Corresponds to amino acid residues A2573 to E2580. The effect of Y2577 phosphorylation is unclear. This is the major in vivo phosphorylation site in NF1 (≥103 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in fish.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NF1 (Neurofibromin) is a large 250-280 kDa cytoplasmic protein acting as a tumour suppressor by regulating cell growth, division, and differentiation. It primarily functions as a GTPase-activating protein (GAP) that inactivates the Ras signalling pathway. It functions as a negative regulator of the RAS/MAPK pathway, accelerating the hydrolysis of Ras-bound GTP, which keeps cells from uncontrolled proliferation. It is a large, multi-domain protein consisting of 2,818 amino acids and functions as a homodimer. It exists in both auto-inhibited (closed) and active (open) conformations. It features a specific domain (GRD) that interacts with Ras, stimulating the hydrolysis of GTP to GDP, turning "on" Ras signals "off". It is involved in additional signaling pathways, including Akt/mTOR and cAMP/PKA, affecting cytoskeleton dynamics and neural development. Besides Ras regulation, it has been shown to interact with proteins like HCN1 to regulate neuronal excitability. While widely expressed, especially in brain, peripheral nerve, lung, colon and muscle, it is most highly concentrated in neurons, Schwann cells, and oligodendrocytes.
