Antibody Immunogen Description: Corresponds to amino acid residues S338 to E344. The effect of T341 phosphorylation is unclear. Phosphorylation at this site in MAT1A has only been reported in one mass spectrometry study. This human phosphosite is highly conserved in vertebrates and also found in yeast. MAT1A is known to be phosphorylated at this site in vitro by PKCa (PRKCA).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MAT1A (Methionine Adenosyltransferase 1A) encodes a crucial liver-specific enzyme that catalyzes the synthesis of S-adenosylmethionine (SAMe) from methionine and ATP. It catalyzes the reaction: L-methionine + ATP + H2O =. S-adenosyl-L-methionine + phosphate + diphosphate. As the main methyl donor in liver cells, this protein exists as tetramers (MAT I) or dimers (MAT III). SAMe is essential for transmethylation reactions, including DNA regulation and neurochemistry. It is exclusively expressed in adult liver and is essential for liver homeostasis; it helps regulate cell proliferation. It forms MAT I (homotetramer) and MAT III (homodimer). Deficiencies in MAT1A causes persistent elevated blood methionine levels (hypermethioninemia).
