Antibody Immunogen Description: Corresponds to amino acid residues H492 to K498. The effect of S495 phosphorylation is unclear. This is the major in vivo phosphorylation site in HMGCS1 (≥55 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and found in fish. HMGCS1 is known to be phosphorylated at this site in vitro by CDK1 (CDC2).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: HMGCS1 (3-Hydroxy-3-methylglutaryl-CoA synthase 1) is a key cytoplasmic enzyme that belongs to the thiolase-like superfamily. In the mevalonate pathway, it catalyzes the rate limiting condensation of acetyl-CoA and acetoacetyl-CoA to produce HMG-CoA with the following reaction: acetoacetyl-CoA + acetyl-CoA + H2O = (3S)-3-hydroxy-3-methylglutaryl-CoA + CoA + H+. HMG-CoA is crucial for cholesterol biosynthesis, and sterol production. It undergoes post-translational modifications such as phosphorylation and ubiquitination. It is often upregulated in various cancers (e.g., gastric, colorectal, lung), HMGCS1 promotes tumour growth, metastasis, and cancer cell survival.
