Antibody Immunogen Description: Corresponds to amino acid residues M85 to T92. S88 phosphorylation inhibits enzyme activity. Phosphorylation at this site in fructose-1,6-bisphosphatase isoform 1 has only been reported in a single mass spectrometry study. However, this human phosphosite is highly conserved in vertebrates and found in fish. FBPase is known to be phosphorylated at this site in vitro by ULK1.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: FBPase (Fructose-1,6-bisphosphatase; FBP1) is a critical, rate-limiting cytosolic enzyme in gluconeogenesis. It catalyzes the irreversible hydrolysis of fructose-1,6-bisphosphate (F1,6BP) to fructose-6-phosphate (F6P) and inorganic phosphate. It plays a crucial role in maintaining blood glucose homeostasis. It plays a role in regulating glucose sensing and insulin secretion of pancreatic beta-cells. It appears to modulate glycerol gluconeogenesis in liver.
