Antibody Immunogen Description: Corresponds to amino acid residues N2276 to Q2282. S2279 phosphorylation stimulates enzyme activity. Phosphorylation at this site in EP300 has only been reported in a single mass spectrometry study. However, this human phosphosite is highly conserved in vertebrates and is found in fish. EP300 is known to be phosphorylated at this site in vitro by ERK2 (MAPK1).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: EP300 (E1A-associated protein p300, also known as p300 or KAT3B) is a large (265 kDa) multifunctional transcriptional coactivator and histone acetyltransferase (HAT) that plays a central role in regulating cell growth, proliferation, differentiation, and apoptosis. It acts as a molecular scaffold to connect transcription factors to the basal transcription machinery. It features a bromodomain, a RING-PHD (CH2) domain, and a catalytic HAT domain. It is closely related to the CREB-binding protein (CBP) and and like CPB can stimulate transcription through activation of CREB. It mediates cAMP-gene regulation by binding specifically to phosphorylated CREB protein. It regulates transcription via chromatin remodeling through acetylation all four core histones in nucleosomes. It functions as a co-activator of HIF1A (hypoxia-inducible factor 1 alpha), and participates in the stimulation of hypoxia-induced genes such as VEGF. It can be methylated at R580 and R604 in the KIX domain by CARM1, which blocks association with CREB, inhibits CREB signaling and activates the apoptotic response. It is also methylated at R2142 by CARM1, which impairs interaction with NCoA2. It is specifically inhibited by the adenovirus oncoprotein E1A.
