Antibody Immunogen Description: Corresponds to amino acid residues E102 to G108. The effect of S105 phosphorylation is unclear. This is the major in vivo phosphorylation site in DNMT3 (≥49 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is not conserved in vertebrates and may be restricted to humans.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: DNMT3A (DNA (cytosine-5)-methyltransferase 3A) is a nuclear DNA methyltransferase that belongs to the class I-like SAM-binding methyltransferase superfamily, C5-methyltransferase family. It features a PWWP domain (targeting chromatin), an ADD domain (Zinc finger for protein interactions), and a C-terminal Methyltransferase (MTase) domain. DNMT3A is required for genome-wide de novo methylation and is essential for the establishment of DNA methylation patterns during development. DNA methylation is coordinated with methylation of histones. It modifies DNA in a non-processive manner and also methylates non-CpG sites. It may preferentially methylate DNA linker between 2 nucleosomal cores and is inhibited by histone H1. It plays a role in paternal and maternal imprinting, and is required for methylation of most imprinted loci in germ cells. It acts as a transcriptional corepressor for ZBTB18. It is recruited to trimethylated 'Lys-36' of histone H3 (H3K36me3) sites. It can actively repress transcription through the recruitment of HDAC activity.
