Antibody Immunogen Description: Corresponds to amino acid residues V485 to P492. T489 phosphorylation induces transcription. This is the major in vivo phosphorylation site in CTNNB1 (≥90 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in sea urchin. CTNNB1 is known to be phosphorylated at this site in vitro by Abl1.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CTNNB1 (Catenin beta-1) is an 88 kDa actin-binding protein and transcription factor that is a regulator of cell adhesion and a key downstream effector in the Wnt signaling pathway. It features consists of an N-terminal domain, a central "armadillo" (ARM) domain composed of 12 repeats that form a rigid, curved structure for binding partners, and a C-terminal domain. In the absence of Wnt, it forms a complex with AXIN1, AXIN2, APC, CSNK1A1 and GSK3B that promotes phosphorylation on N-terminal Ser and Thr residues and ubiquitination of CTNNB1 via BTRC and its subsequent degradation by the proteasome. In the presence of Wnt ligand, CTNNB1 is not ubiquitinated and accumulates in the nucleus, where it acts as a coactivator for transcription factors of the TCF/LEF family, allowing activation of Wnt responsive genes, including c-Myc an Cyclin D1. It also acts as a coactivator for other transcription factors, such as NR5A2. It is involved in the regulation of cell adhesion, as component of an E-cadherin:catenin adhesion complex. It also functions as a negative regulator of centrosome cohesion. It is also implicated early embryonic development and tumorigenesis.
