Antibody Immunogen Description: Corresponds to amino acid residues V353 to Q359. The effect of T356 phosphorylation is unclear. Phosphorylation of this site in ATASE has been reported in only one mass spectrometry study. This human phosphosite is highly conserved in vertebrates and also found in yeast.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ATASE (Amidophosphoribosyltransferase; GPAT) is a 57 kDa metabolic enzyme that carries out the first committed, rate-limiting step in de novo purine nucleotide biosynthesis. ATASE catalyzes the reversible formation of 5-phosphoribosyl-1-amine (PRA) from phosphoribosylpyrophosphate (PRPP) and glutamine in the reversible reaction: 5-phospho-beta-D-ribosylamine + L-glutamate + diphosphate <=> 5-phospho-alpha-D-ribose 1-diphosphate + L-glutamine + H2O. PRA is essential for DNA/RNA synthesis and cell proliferation. The active form is often a tetramer (sometimes referred to as a "doughnut-shaped" molecule) consisting of subunits organized into two domains. The protein binds [4Fe-4S] clusters (one per subunit) and requires Mg2+ (or Mn2+) for activity. High levels of PRPP activate ATASE by triggering a structural change that lowers the Km for glutamine by 100-fold. It is inhibited by downstream purine nucleotides, specifically AMP, GMP, and IMP.
