Antibody Immunogen Description: Corresponds to amino acid residues I171 to E183. The effects of T174 and S180 phosphorylation are unclear. These are two of the minor in vivo phosphorylation sites in arginosuccinate synthase (≥9 and >8 reports, respectively, by mass spectrometry in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in yeast.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ASS1 (Argininosuccinate synthase) belongs to the argininosuccinate synthase family. Type 1 subfamily. It is one of the enzymes of the urea cycle, which is the metabolic pathway that transforms neurotoxic ammonia produced by protein catabolism into inocuous urea in the liver of ureotelic animals. It catalyzes the formation of arginosuccinate from aspartate, citrulline and ATP in the reaction: L-citrulline + L-aspartate + ATP => 2-(N(omega)-L-arginino)succinate + AMP + diphosphate (PPi) + H+. Together with ASL, it is responsible for the biosynthesis of arginine in most body tissues. It exists as a homotetrameric protein, composed of 45-kD monomers, which stabilizes the active site and improves catalytic efficiency. It is highly expressed in adult liver and kidney. Deficiency of ASS1 causes Citrullinemia Type I, an autosomal recessive urea cycle disorder. It is often downregulated (silenced) in various cancers (e.g., sarcomas, bladder cancer), causing arginine auxotrophy, making these cells susceptible to arginine deprivation therapy.
