Antibody Immunogen Description: Corresponds to amino acid residues R269 to Q275. S272 phosphorylation stimulates enzymatic activity and import to the nucleus. This is a minor in vivo phosphorylation sites in ALOX5 (≥2 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and found in fish. ALOX5 is known to be phosphorylated at this site in vitro by CaMK2a, MAPKAPK2, and PKACa.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ALOX5 (polyunsaturated fatty acid 5-lipoxygenase) catalyzes the oxygenation of arachidonate ((5Z,8Z,11Z,14Z)-eicosatetraenoate) to 5-hydroperoxyeicosatetraenoate (5-HPETE) followed by the dehydration to 5,6- epoxyeicosatetraenoate (Leukotriene A4/LTA4),. These are the first two steps in the biosynthesis of leukotrienes, which are potent mediators of inflammation. It also catalyzes the oxygenation of arachidonate to produce 8-hydroperoxyicosatetraenoate (8-HPETE) and 12-hydroperoxyicosatetraenoate (12-HPETE), as well as has lipoxin synthase activity to convert (15S)-HETE into a conjugate tetraene. The preferred substrate of ALOX5 is arachidonate, but it can also metabolize oxidized fatty acids derived from arachidonate such as (15S)-HETE, eicosapentaenoate (EPA) such as (18R)- and (18S)-HEPE or docosahexaenoate (DHA) which lead to the formation of specialized pro-resolving mediators (SPM) lipoxin and resolvins E and D, respectively, so it can participate in generation of anti-inflammatory responses.
