Antibody Immunogen Description: Corresponds to amino acid residues I803 to G810. The effect of S806 phosphorylation is unclear. This is phosphorylation site in ACO1 has only been reported in one mass spectrometry study in PhosphoSitePlus. This human phosphosite is highly conserved in vertebrates and found in fish.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ACO1 (aconitate hydrolase) is a 98-100 kDa cytoplasmic, bifunctional iron sensor that switches between two activities depending on iron availability. When iron is abundant, it binds a 4Fe-4S cluster and acts as a cytoplasmic aconitase. Under iron deprivation, it promotes its mRNA binding activity through which it regulates the expression of genes involved in iron uptake, sequestration and utilization. It binds to iron-responsive elements (IRES) in the 5' or 3' untranslated region of target mRNAs to prevent the translation of ferritin and aminolevulinic acid synthase and stabilizing the transferrin receptor mRNA. It converts citrate to D-threo-isocitrate. Aberrant ACO1 function is linked to iron disorders, anemia, and neurodegenerative conditions, with variants associated with hemoglobin concentrations.
