Antibody Immunogen Description: Corresponds to amino acid residues I1339 to D1345. The effect of T1342 phosphorylation is unclear. This is the major site of phosphorylation in ACC2 by mass spectrometry (≥30 reports). This human phosphosite is highly conserved in mammals and also found in the platypus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ACACB (acetyl-CoA carboxylase-2; ACC2) is a mitochondrial enzyme that catalyzes the carboxylation of acetyl-CoA to malonyl-CoA ((ATP + acetyl-CoA + HCO3- <=> ADP + phosphatse + malonyl-CoA)) and plays a central role in fatty acid metabolism, the first and rate-limiting step of de novo fatty acid biosynthesis. The two step reaction starts with the ATP-dependent carboxylation of the biotin carried by the biotin carboxyl carrier (BCC) domain followed by the transfer of the carboxyl group from carboxylated biotin to acetyl-CoA. It features a central biotin carboxyl carrier (BCC) domain, an N-terminal biotin carboxylase (BC) domain, and a C-terminal carboxyl transferase (CT) domain. Through the production of malonyl-CoA that allosterically inhibits carnitine palmitoyltransferase 1 at the mitochondria, it is thought to negatively regulate fatty acid oxidation. Unlike its cytosolic isoform ACACA, ACACB is located on the outer mitochondrial membrane, anchored by a unique N-terminal sequence. Along with its cytosolic isozyme ACACA, it promotes lipid storage.
