Antibody Immunogen Description: Corresponds to amino acid residues G429 to L436. The effect of Y433 phosphorylation is unclear. This is the major site of in vivo phosphorylation of STAG2 (≥42 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). This human phosphosite is highly conserved in vertebrates and also found in the frog.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STAG2 (Stromal Antigen 2) is a nuclear core subunit of the cohesin complex (along with SMC1, SMC3, and RAD21) and belongs to the SCC3 family. It is essential for sister chromatid cohesion during cell division, after DNA replication, DNA damage repair, and gene expression regulation. The cohesin complex forms a large proteinaceous ring within which sister chromatids can be trapped. At anaphase, the complex is cleaved and dissociates from chromatin, allowing sister chromatids to segregate. The cohesin complex may also play a role in spindle pole assembly during mitosis. As a crucial component of mitosis and meiosis, it ensures proper chromosome segregation. Mutations in STAG2 are associated with various cancers (e.g., bladder cancer, AML) and developmental disorders. While similar to STAG1, STAG2 has distinct roles in 3D genome organization in the mitotic cohesin complex, such as regulating chromatin loops.
