Antibody Immunogen Description: Corresponds to amino acid residues D607 to P616. The effects of S610 and S613 phosphorylation are unclear. These are the major sites of in vivo phosphorylation based on ≥56 and ≥69 mass spectrometry reports, respectively, in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SMARCA4 (Transcription activator BRG1) is a 185–205 kDa ATP-dependent catalytic subunit of the SWI/SNF complex, which remodels chromatin to regulate gene expression in the nucleus of cells. It belongs to the neural progenitor-specific and the neuron-specific chromatin remodeling complexes (npBAF and nBAF, respectively). As a core ATPase subunit, it uses ATP to alter nucleosome structure, facilitating DNA transcription and repair and cell cycle control and differentiation. It Interacts with p53 and the androgen receptor, regulating gene expression to prevent uncontrolled cell growth. It is a component of remodeling complexes including Swi/Snf-A (BAF), Swi/Snf-B (PBAF), Brm, WINAC and Brg1. Each complex contains a catalytic subunit (either SMARCA4 or SMARCA2), and at least SMARCE1, BAF53A or BAF53B, SMARCC2 and SMARCB1. SWI/SNF complexes are required for mammalian development and are mutated in ~20% of all human primary tumours. It is a transcriptional coactivator of nuclear hormone receptors. It colocalizes with ZEB1 in stroma of normal colon as well as in de-differentiated epithelial cells at the invasion front of colorectal carcinomas.
