Antibody Immunogen Description: Corresponds to amino acid residues G510 to Q516. The effect of phosphorylation of Y513 is unclear, and it is a minor site of phosphorylation (≥1 report by mass spectrometry in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Smad4 (Mothers against decapentaplegic homolog 4; DCP4) is a tumour suppressor protein and transcription factor that belongs to the dwarfin/SMAD family. It functions as the "common-partner" SMAD (co-Smad) in the TGF-β and BMP signaling pathways, mediating cell proliferation, differentiation, and apoptosis by transporting activated receptor-Smads into the nucleus. It forms complexes with receptor-activated Smads (like Smad2/3) to enter the nucleus and bind to DNA. When recruited by MSTN, it promotes atrophy response via phosphorylated SMAD2/4. MSTN decrease causes SMAD4 release and subsequent recruitment by the BMP pathway to promote hypertrophy via phosphorylated SMAD1/5/8. It acts synergistically with SMAD1 and YY1 in bone morphogenetic protein (BMP)-mediated cardiac-specific gene expression.
